It was built to a design by architect Ennio Canino between 1970 and 1972, and was solemnly consecrated on 29 May 1972 by Cardinal Vicar Angelo Dell'Acqua. The church was visited by two popes: Paul VI in April 1973, and John Paul II in January 1984.
The interior houses a bronze Madonna by Perrotta, and a Screaming Crucifix, also in bronze, by U. Montalbano; the crypt displays a series of mosaics depicting the patron saints of the countries of origin of the Giuliano-Dalmatian refugees (Upper East Adriatic). A statue of St Mark's lion, donated by a noble family in the 1970s, has stood on the boat at the central entrance to the church since December 2016. The year 2017 saw the complete renovation of the interior flooring of the church in Persian travertine.
The church is a parish seat, erected on 9 March 1950 by decree of Cardinal Vicar Francesco Marchetti Selvaggiani Pastoris vigilantis. Since 1973 it has been the seat of the cardinal title of San Marco in Agro Laurentino. It was entrusted to the care of the Friars Minor Conventual until 31 August 2016. Since 1 September of the same year, it has been entrusted to the care of the diocesan clergy.
On the original site where the church stood, there are gardens named after Maestro Lodovico Zeriav. Within them is the monument of the Roman She-wolf of Pula.
